Nickel oxide nanoparticles induced DNA damages in human liver cells

Background and Aims: Nickel oxide nanoparticles (NiO-NPs) are one of the most used nanoparticles, especially as photosensitizers. Although some studies evaluate their toxicity in liver, information about at cellular molecular levels is still controversial. In present study, it was aimed to investigate vitro toxic potentials NiO-NPs (average size 15.0 nm) liver (HepG2) cell line. Methods: were characterized by Transmission Electron Microscopy (TEM), uptake NPs morphologic changes evaluated TEM Inductively Coupled Plasma-Mass Spectrometry (ICP-MS), cytotoxicity MTT neutral red (NRU) tests, comet assay for genotoxicity, Annexin V-FITC/propidium iodide (PI) apoptosis detection kit apoptosis/ necrosis evaluation Enzyme-Linked Immune Sorbent Assays (ELISA) kits potential oxidative damage. Results: Our results showed that led morphological cells, caused death (IC50 146.7 µg/mL MTT) mainly apoptosis. Genotoxicity damage observed be a dosedependent manner. Conclusion: Results confirm previous data draw attention effects NiO-NPs; further vivo studies need done clarify safety or NiO-NPs.
